Research Overview
FOXO4-DRI is a synthetic D-retro-inverso peptide that disrupts the interaction between FOXO4 and p53, two proteins that cooperate to keep senescent ("zombie") cells alive. Senescent cells are damaged cells that stop dividing but refuse to die, accumulating with age and secreting inflammatory factors (the SASP - senescence-associated secretory phenotype) that damage surrounding tissue.
By disrupting FOXO4-p53 binding, FOXO4-DRI forces senescent cells into apoptosis (programmed cell death) while sparing healthy cells. This makes it one of the first true "senolytics" - drugs that selectively kill senescent cells. In aged mice, FOXO4-DRI restored fur density, renal function, and fitness levels.
